چکیده

Gene bank accessions are necessary for implementing many research and breeding projects. However, a great number of contaminated or confused. If such used, the results obtained from these projects inaccurate non-reproducible. There methods that allow almost perfect genotype identification; nevertheless, they relatively recent cannot be compared with characteristics original accessions. Growing resistant cultivars is an environmentally safe cheap way disease management knowledge diverse resistance genes their combinations can used to identify varieties verify authenticity homogeneity. For this purpose, all 172 core collection (CC) Czech winter barley (Hordeum vulgare) gene bank, originating 35 countries, were studied. tests, 51 reference isolates Blumeria graminis f. sp. Hordei, collected in nonpolar continents over period 63 years representing global virulence/avirulence diversity pathogen, used. Only 25 homogeneous (genetically uniform), whereas 147 heterogeneous due presence different genotypes. In total, 17 found singly combinations; 76.3% identified carried alleles at Mla locus. To purify CC, progenies individual plants must multiplied homogeneity seed should confirmed subsequently studied more advanced methods.

Heterogeneity of Powdery Mildew Resistance Revealed in Accessions of the ICARDA Wild Barley Collection

The primary genepool of barley comprises two subspecies - wild barley (Hordeum vulgare subsp. spontaneum) and cultivated barley H. vulgare. subsp. vulgare. The former originated 5.5 million years ago in southwest Asia and is the immediate ancestor of cultivated barley, which arose around 10,000 years ago. In this study, the specific resistance of a set of 146 wild barley accessions, maintained ...

Allele Mining in Barley Genetic Resources Reveals Genes of Race-Non-Specific Powdery Mildew Resistance

Race-non-specific, or quantitative, pathogen resistance is of high importance to plant breeders due to its expected durability. However, it is usually controlled by multiple quantitative trait loci (QTL) and therefore difficult to handle in practice. Knowing the genes that underlie race-non-specific resistance (NR) would allow its exploitation in a more targeted manner. Here, we performed an as...

Virus-induced gene silencing-based functional characterization of genes associated with powdery mildew resistance in barley.

We successfully implemented virus-induced gene silencing (VIGS) in barley (Hordeum vulgare) for the functional characterization of genes required for Mla13-mediated resistance toward the biotrophic barley pathogen Blumeria graminis f. sp. hordei. Initially, barley cultivars were screened for their ability to host the barley stripe mosaic virus (BSMV)-VIGS vector by allowing its replication and ...

Identification of Genes Required for the Function of Non-Race-Specific mlo Resistance to Powdery Mildew in Barley.

Recessive alleles (mlo) of the Mlo locus in barley mediate a broad, non-race-specific resistance reaction to the powdery mildew fungus Erysiphe graminis f sp hordei. A mutational approach was used to identify genes that are required for the function of mlo. Six susceptible M2 individuals were isolated after inoculation with the fungal isolate K1 from chemically mutagenized seed carrying the mlo...
